A synthetic matrix that supports primary satellite cell survival, proliferation, and differentiation has been engineered. We further develop an injection-free strategy to facilitate the delivery of the satellite cells to the dystrophic diaphragm. Transplanted satellite cells using the hydrogel exhibit superior retention, and successfully engraft and differentiate to form de novo muscle fibers in the dystrophic diaphragm.
