In the study, using a cold cathode fluorescent lamp with low heat, long lifetime, low light failure and good color rendering index as the microscope light source, and using contrast of color peak signal-to-noise ratio (CPSNR) and structural similarity index (SSIM) to compare the microscope images of a rat kidney slice with halogen lamp. The CRI values of a halogen lamp and the CCFL measured by an integrating sphere were 98.6, 85.2, respectively. The images yielded from both the halogen lamp and the CCFL as the light sources had a CPSNR of 26.83 dB and a SSIM of 0.9148. Even if the image from CCFL light source have a little distortion, but the similarity of two images between the halogen and CCFL light source as high as 90%. CCFL has been confirmed to provide microscopic lighting successfully.
