Conclusions: In agreement with our previous results, continuous IL-4 delivery was effective in preventing particle induced osteolysis both after 4 and 8 weeks of combined particle and IL-4 delivery. Furthermore, delayed IL-4 treatment was not only effective in preventing further UHMWPE particle induced bone loss but also in repairing the bone loss present at the beginning of the treatment. Results demonstrate the translational potential of this treatment strategy targeting local macrophage polarization.
