The results indicate that using click chemistry to attach EpCAM to the surface of alkyne-functionalized PLGA particles is a viable surface modification route. There was a significant difference of about 10% in the uptake of EpCAM modified particles by EpCAM+ cells relative to all the cancer cells, indicating that targeting to EpCAM is occurring, though could be improved upon. Future studies include decreasing the incubation time to limit the effect of nonspecific uptake as well as exploring other targeting ligands for more specific uptake. This work shows promise towards targeting the difficult to target CSCs in breast cancer.
