Covalent conjugation of siRNA to hydrogels allows temporal and spatial control over its release via hydrolytic degradation of the linkers between the siRNA and the hydrogels. The released siRNA was bioactive and could transfect HeLa cells without the need of a transfection reagent. This approach provides a promising strategy for temporally and spatially controlling siRNA release for tissue engineering and disease therapeutics.
