In this study, we successfully developed new biodegradable stents with trilayered architecture and demonstrated their capability to provide bilateral release of an antiproliferative drug PTX and an antibiotic drug OFLX. The release of PTX and OFLX were sustained for months, with highly adjustable release profiles and rates by changing the initial drug loading contents or adding PEG in the films. Bacterial adherence to the OFLX layer of film was substantially inhibited throughout the 31-day investigation period; meanwhile, the films suppressed the proliferation and growth of PC-3 cells over the elution period of PTX. With the advantage of co-delivering and releasing dual drugs in a bilateral manner, the trilayered stents showed great potential for combinatorial treatments of complex endoluminal restenosis.
