In this study, we demonstrated that immobilization of biomolecules via layer-by-layer (LBL) polyelectrolyte deposition is a convenient way to modulate cell behavior. Biomolecules are first conjugated onto polyelectrolytes and then deposited onto substrates via the deposition of polyelectrolyte multilayer (PEM) films. We showed that deposition of cell adhesive molecules (e.g., RGD) enhanced the adhesion, proliferation and mineralization of osteoblasts, while conjugation of non-fouling materials (e.g., PEG) inhibits cell adhesion. PEM combined with photolithography can be used to prepare micropatterned surfaces for cell patterning and co-culture.
