Acellular buffalo small intestinal submucosa and fish swim bladder for repair of full-thickness skin wounds in rabbit

In the present study acellular matrix from small intestine of water buffalo (Bubalus bubalis) and swim bladder of fresh water fish (Labeo rohita) were prepared using ionic biological detergent. The small intestine treated with 2% ionic biological detergent for 36h under constant agitation showed complete loss of cellularity. The mucosa was completely acellular, submucosa had normal arrangement of collagen fibres, and muscular layer was acellular with normal muscle fibre structure. Histological examination confirmed the acellularity of the matrices. The nucleus and cytoplasmic cellular components were successfully removed with no evidence of remaining cells, nuclei or other cell fragments and resulted into acellular intestinal matrix consisting of primarily collagen with good tensile strength. The swim bladder treated with 0.5% ionic biological detergent for 24h under constant agitation showed complete loss of cellularity. The tunica externa and interna were completely acellular. Contrary to native tissue the collagen fibres were loosely arranged. Acellular swim bladder was cross-linked with 1,4-butanediol diglycidyl ether (BDDGE) for 48 h and acellular intestinal matrix was cross-linked with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) for 48 h.
