Overall, exposure to a collagen matrix, which physically resembles the skeletal muscle's extracellular matrix, induces rapid growth, maturation and function of SC-derived myotubes. In the presence of necrotic myocytes, the collagen matrix's ability to augment mature myotube formation is greatly increased. In vivo, application of the collagen matrix reduced the loss of necrotic muscle function, but no functional change was observed in the atrophic model. This was echoed by transcription profiles which showed that regeneration was evident only in the necrotic mdx muscle. Together, these results suggest that application of a collagen-based matrix has the potential to augment skeletal muscle regeneration and that its regenerative abilities may be more successful for treating necrotic, but not atrophic muscle.
