Delivery of mRNA by a Novel pH-Responsive Triblock Copolymer

We have developed and characterized a series of triblock copolymers with pH-responsive endosomolytic behavior capable of mediating mRNA transfection in a macrophage cell line in vitro. Preliminary evidence indicates that these copolymer systems are superior mRNA transfection vectors compared to PEI and can be optimized to be comparable to the commerical standard Lipofectamine. These results demonstrate that this system is a promising polymeric delivery vector for mRNA-based gene therapy applications. Ongoing studies seek to improve biocompatibility and polyplex stability through modifications of the PEGMA block.
