The findings of this work suggest that MCM may aid in supporting VSMC attachment. Furthermore, while calponin expression by VSMCs in MCM increased over time, this effect was not seen in co-culture, suggesting that cell-cell contact, rather than secreted factors, may contribute to the modulation of calponin in co-culture. The fact that the MCM and co-culture systems both enhanced cell migration into the scaffold indicates a key role for monocytes and their secreted factors in this process. Ongoing work is assessing the influence of growth factors (TGF-β1, PDGF-BB) on VSMC response, while also looking at other secreted factors from monocytes on D-PHI, in order to aid in explaining the mechanism of action of monocytes in VSMC migration into the scaffold and the modulation of their contractile state.
