Experiments were conducted with leaves as explants collected from the test-tube plantlet of Pacific Gala to study the effects of different concentrations of NAA and TDZ,combination of 6-BA and TDZ on adventitious shoot regeneration in vitro.The results showed that with the increase of concentration of TDZ,the regeneration rates and regenerated adventitious shoots number increased at first and then decreased in the medium with NAA and TDZ.Under the condition of 2.0 mg·L-1 TDZ,regeneration rates reached 100%,and the maximum adventitious shoots number per explants was 15.8,which decreased 6.2 or 8.5 respectively when supplemented 2.0 mg·L-1 or 4.0 mg·L-l 6-BA to the medium of MS + 0.5 mg·L-1 NAA + 2.0 mg·L-1 TDZ.The regeneration rates of faced up leaves were 2.2% more than that of faced down leaves,while the number of adventitious shoots per explants was more than 2.44 correspondingly.The optimum medium for Pacific Gala regeneration in vitro was MS + 0.5 mg·L-1 NAA + 2 mg·L-1 TDZ.
