Development of Liposomal DNA Repairing Enzymes for Topical Delivery

A liposomal delivery system is used to encapsulate a DNA repairing enzyme T4 endonuclease V (T4N5), which repairs UV-induced cyclobutane pyrimidine dimers in DNA, to increase the penetration to the skin, thereby enhancing therapeutic effects. The concentration of T4N5 liposome is 176 ng/ul and the particle size is approximately 100nm. A high encapsulation efficiency of T4N5 liposome is up to 65%. In vitro efficacy test where nude mice skin exposed to UV light treated with T4N5 liposome shows effectively decreased the number of T-T dimers, thereby enhancing skin repair at 6 and 24 hours. When used continuously, the effect is reinforced. This presentation will focus on the formulation and the in vitro and in vivo studies.
