The objectives of this study were to correlate biopharmaceutical properties and negative food effects (NFEs) of drugs and develop in vitro/in situ models to predict NFEs. Data analysis shows that food effects correlate primarily to drugs' dissociation constants and Caco-2 permeability. Rat intestinal permeability from SPIP studies at pH 5 and 6.7 can be used to simulate NFEs. Therefore, in situ rat intestinal model can be used to predict NFEs.
