Ketorolac fatty acid esters were synthesized; evaluated for lipophilicity, chemical/enzymatic hydrolysis in skin/liver homogenates and plasma, and skin permeation/accumulation profiles in propylene glycol pretreated intact, stratum corneum stripped and delipidized hairless mouse skins. Esters were highly lipophilic, chemically stable, enzymatically unstable, impermeable into lipophilic receptor solution with varied dermal accumulation.
