Proteomics of barley grain – a new tool for selection of malting cultivars

The knowledge of the genotype alone does not permit to select better malting cultivars. To this aim, detailed elucidation of the protein composition is necessary. Identification of proteins extracted from barley grains by the proteomic strategy based on a combination of gel electrophoresis, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and bioinformatics was performed. Simplification of extracted protein mixtures was achieved by sequential extractions with various solvents based on modified Osborne system. Several proteins were identified in this way. It seems that this approach can become an important tool for proteomics of barley. The detailed knowledge of the grain protein composition will help to determine the environmental effects on grain quality and produce further insight into the relationships between protein composition and the technological properties of malting barley.
