A number of hydrotropic agents were evaluated for increasing water solubility of paclitaxel. N,N-Diethylnicotinamide was the most effective hydrotropic agent in our study. In general, the presence of a pyridine or benzene ring was required for good hydrotropy. The effects of various hydrotropic agents were analyzed for their structure-activity relationships. This information obtained for the chemical structures of paclitaxel hydrotropy can be used for preparing hydrotropic polymers for paclitaxel. In addition, the information may be used as a basis for examining hydrotropic properties for other poorly soluble drugs.
