The uptake and intracellular trafficking of three non-viral gene delivery systems, polyethylenimine, chitosan, and Lipofectamine 2000 were evaluated in an in vitro dissociated dorsal root ganglion (DRG) culture. All formulations were rapidly taken up by the neurons but did not traffic through acidic vesicles. A significant barrier to gene transfer in DRG neurons appears to be the release of plasmid DNA from the complexes into the cytosol.
