Preparative or large scale liquid chromatogrpahy has been widely applied as a separation technique for the production of high-purity drugs, biomolecules and carbohydrate. It is not only feasible for muticomponet separation but also simple in operation and equipment. However, a conventional single-run elution chromatogrpahy has a disadvantag ethat its production rate is lower. Wankat~(1) proposed several concepts to overcome the drawback. Barker and Ganetsos ~(2) made the comparison between several techniques of countercurrent and crosscurrent chromatography. Ruthven and Ching~(3) have reviewed the simulated countercurrent or moving-bed principle. Simulated moving bed chromatogrpahy is the process of gaining higher production rate but is sophisticated technique.
