The in vitro ~(14)C-labelled kaft pulp assay as used for evaluation of laccases and mediators intended for pulp bleaching

In the presence of K-ferricyanide as oxidant at pH 10, ~(14)C-ring-ablelled synthetic lignin (DHP) was covalently coupled to unbleached kraft pulp. This chemically labelled kraft pulp (CLKP) was used in a small scale (30 mg pulp in 1.5 ml buffer) for testing ligninolytic enzymes and mediators for possible use in pulp bleaching. The results from this assay correlated well with laccase/mediator kraft pulp delignification plus TCF-bleaching.
