A simulation study on the fractionation of recombinant protein inclusion bodies from cell debris in a centrifuge is presented. This study is based on recently-developed E. coli debris comminution model coupled with experimetnally-determined "grade-efficiency" curves for inclusion odies and cell debris in a disc-stack centrifuge. The efffect of homogenisation and centrifugation conditions on inclusion-body recovery and purity is simulated and discussed. Different centrifugation strategies such as repeated centrifugation and the interspesion of homogenisation between centrifuge passes are also discussed.
