The microsatellite DNA from seven main fresh water aquatic species including common carp, silver crucian carp, grass carp, silver carp, bighead carp, snout bream, Chinese river crab were cloned by two methods. One method is screening short inserted DNA library using a few synthetic oligo-nucleotide simple repeated sequence labeled at the 5'end with [γ-p~(32)] ATP. The other method is based on using biotinylated probes attached to magnetic beads to isolate pieces of DNA containing the microsatellites. We got 1180 microsatellite DNA for common carp, 137 for silver crucian carp, 114 for grass carp, 138 for silver carp,75 for bighead carp, 108 for Chinese river crab, 120 for snout bream. PCR primers were designed using soft ware PRIMER5. A total of 210 primer sets were tested for polymorphism on each species panels of 6 animals. 60% markers were found to be polymorphic. These polymorphic markers as well as non-polymorphic markers will be very valuable as genetic tools for using in strain identification, parentage analysis, quantity trait loci analysis, population structure analysis and marker-based selection studies.
