The requirement for selective cellular adhesion, proliferation and migration in biomedical applications requires integrative understanding on cell-biomaterials interaction. Hence cell response to chemical, mechanical and topological properties of substrate has been intensively studied. However, the effect of cell seeding density on cell-biomaterials interaction in in-vitro study was seldom considered. This work demonstrated the important roles of cell seeding density in cell-biomaterials interaction by studying cyto-response to macromolecules modified poly (L-lactic acid) (PLLA) using two seeding densities, low seeding density of 5000 cells/cm2 and high seeding density 50,000 cells/cm2. For the first time, cell-biomaterials interaction was divided into two stages, the early stage within first several hours after cell seeding where cell adhesion dominates, and the later stage after 6 hr of cell seeding where cell proliferation dominates.
