A dual modality of free space fluorescence diffuse optical tomography and ultrasound imaging was demonstrated for the first time. The design combined a laser source with electron-multiplying charge-coupled device (EMCCD) on a rotation gantry with the structural information provided by an ultrasound transducer to reconstruct fluorescence diffuse optical tomography images. Optical data were obtained using a 660 nm laser at 16 angles in free space, and the fluorescence was in trans-illumination mode. In this work, a small feasibility study was performed to demonstrate that the fluorophore distribution could be recovered accurately and quantitatively using this imaging system.
