The aim of this study was to investigate the mucoadhesion of poly(acrylic acid) and thiolated poly(acrylic acid)-cysteine matrix tablets for gastroretentive delivery systems using riboflavin as a model drug. For this aim, three different in vitro and in vivo methods were used for the comparison of mucoadhesion. For in vitro mucoadhesion studies, two methods, namely rotating cylinder method and Texture Profile Analysis were used. In rotating cylinder method, freshly excised porcine stomach mucosa was used. On the other hand, mucin discs were employed as the model substrate in Texture Profile Analysis. The results of both methods showed that mucoadhesion of thiolated tablets were higher than unmodified ones. Thereafter, the in vivo efficacy of thiomers was evaluated with minitablets in rats. In vivo studies demonstrated the gastroretention time of thiolated tablets up to 6 hours. Consequently, the study suggests that poly(acrylic acid)-cysteine matrix tablets might be promising formulation for gastroretentive delivery systems.
