We developed a novel class of biodegradable particles based on lipids for drug and vaccine delivery. Interbilayer-crosslinked multilamellar vesicles (ICMVs) were synthesized by fusing liposomes into multilamellar vesicles, followed by the formation of crosslinks between headgroups of apposed bilayers within the multilamellar structures. The resulting ICMVs exhibited colloidal stability with particle diameter of 250 ± 40 nm, and greater than 70% encapsulation efficiency (407.5 + 18.7 ug ova per mg of lipid) was achieved in these particles with ovalbumin used as a model macromolecule. We demonstrate in vivo application of these new materials in vaccine development.
