Rumex acetosa L. is one of the best-studied dioecious plant species in cytoge-netics. Female and male plants have different sex-chromosome complements XX and and XY_1Y_2, respectively. To develope the chromosome specific molecular markers, autosomes and sex-chromosomes (X, Y_1, and Y_2) were isolated using micromanipulation and amplified with degenerated oligonucleotide primed PCR (DOP-PCR). The PCR-cloned fragments were subjected to an amplified fragment length polymorphism (AFLP) analysis. We obtained over five hundreds of X and/or Y chromosome specific bands, ranging from 210 to 600bp in size. Some of them were Y chromosome specific.
