A genome-scale metabolic network of C. Acetobutylicum ATCC 824 comprising 502 reactions and 479 metabolites was reconstructed from various annotation data. Using this model, we compared the simulation result with fermentation data. We successfully predicted the fluxes of acidogenic phase using typical flux balance analysis (FBA), and the fluxes of solventogenic phase was also calculated using nonlinear programming which assumes the minimal fluctuation of fluxes except acid and solvent production. In addition, essential genes were predicted via single gene deletion studies. The genome-scale model of C. Acetobutylicum can be used for genome wide analysis as well as for designing of strain development strategy for improving production of biochemicals including 1-butanol.
