Provided is a tumor immunotherapy method based on the activation of an NK cell. Further provided is a use of nucleobindin (NUCB) or a promoter thereof in the preparation of a drug or a preparation for promoting the proliferation of NK cells, treating tumors, and/or killing or inhibiting tumor cells. The NK cell is activated by means of the method, and the obtained NK cell has a broad-spectrum anti-tumor effect without the identification of a tumor-specific antigen, such that the priming of an immune system is fast.
