Methods, assays, and kits are disclosed for identifying ligands for biological molecules, such as proteins, lipids, carbohydrates, or nucleic acids. The disclosed method can be a quantitative binding assay for a target, which avoids the challenges of target purification and can provide a systematic approach for discovering and targeting allosteric binding sites.
