The present disclosure relates to peptide immunogen constructs targeting portions of calcitonin gene-related peptide (CGRP), compositions containing the constructs, antibodies and constructs induced by the constructs, and methods of making and using compositions thereof will be. The disclosed peptide immunogen constructs have greater than about 30 amino acids and contain (a) greater than about 7 contiguous amino acid residues from the CGRP receptor binding or activation region of the full-length CGRP protein to a B cell epitope; (b) a heterologous Th epitope; and (c) an optional heterologous spacer. The disclosed CGRP peptide immunogen constructs stimulate the production of highly specific antibodies to CGRP for the prevention and/or treatment of migraine.
