Disclosed is a method for separating mannitol, adenosine and ergosterol from a sample to be tested. The method comprises: subjecting a sample to be tested to an elution treatment sequentially with a first eluent, a second eluent, a third eluent, and a fourth eluent in a solid phase extraction column, and respectively collecting an elution product of the first eluent, an elution product of the second eluent, and an elution product of the fourth eluent so as to sequentially obtain eluents containing mannitol, adenosine and ergosterol, wherein the first eluent, the second eluent, the third eluent, and the fourth eluent are each independently a methanol aqueous solution, or an ethanol aqueous solution, or an acetonitrile aqueous solution, or any combination of the three. The method can achieve the simultaneous purification of adenosine and ergosterol from the sample to be tested, and the purities of the obtained adenosine and ergosterol are greatly improved.
