A content determination method for enhancing immunoturbidimetry on the basis of plasma optical nanopores, comprising: step 1: mixing a first reagent with a sample to be determined, and adding the mixture into a porous microwell plate in which a nano optical sensor chip is provided; step 2: measuring a first turbidity change curve of a mixture reaction system in the porous microporous plate within a pre-determined wavelength range by using an enzyme-labeling instrument; step 3: adding a second reagent into the solution in the porous microwell plate; step 4: measuring a second turbidity change curve of the mixture reaction system in step 3 in the pre-determined wavelength range by using the enzyme-labeling instrument; step 5: obtaining a standard curve representing the relationship between the content of a standard substance to be detected in the first reagent and the second reagent and the change in turbidity; and step 6: comparing the first turbidity change curve obtained in step 2 and the second turbidity change curve obtained in step 4 with a standard curve so as to obtain the content of the substance to be detected.
