Systems and methods for evaluating a biological sample are described herein. The method includes: characterizing the pattern with speckle to be applied by the diffuser; positioning the biological sample relative to the at least one coherent light source such that the at least one coherent light source illuminates the biological sample; diffusing light generated by the at least one coherent light source; capturing a plurality of illuminated images having an embedded speckle pattern of the biological sample based on the diffused light; iteratively reconstructing an illuminated image with a plurality of speckles of a biological sample to recover an image stack of the reconstructed image; stitching together each image in the image stack to create an entire slide image, wherein each image in the image stack at least partially overlaps a neighboring image; and identifying one or more features of the biological sample. The method may be performed by a near-field Fourier tychographic system.
