The present invention relates to a method for performing cell-free protein synthesis or other fluorescent assays in the context of cell-free protein synthesis and a multi-well plate for performing the method. The method includes providing a multi-well plate, the plate comprising a base and a cover, wherein the base includes multiple wells, each well comprising one or more sidewalls, a bottom, and an opening, wherein the cover is configured to close the openings of the wells, and wherein each of the multiple wells has a volume of 20 µL or less. A volume of fluid is provided in at least some of the wells, wherein the fluid comprises a cell-free reaction mix and/or a fluorescent assay. Reagents comprising biochemical factors, at least one of template DNA, template RNA, additives and reaction co-factors are introduced into the wells. A cover is placed on top of the base to close the openings of the wells. The fluid is in contact with both the well bottom and the cover. The wells are incubated for a certain period of time.
