Disclosed in the present invention is a detection method based on supercritical fluid chromatography and ionic liquid post-column addition reaction, comprising the following steps: (1) using n-heptanol, tetrahydrofuran and water to prepare a supermolecular solvent; (2) sample pretreatment: using the supermolecular solvent to extract a sample so as to prepare a liquid to be detected; and (3) detecting perfluorinated compounds in the liquid using a method combining supercritical fluid chromatographic separation, dual-cation type ionic liquid chromatographic post-column addition reaction and electrospray mass spectrometry detection. The present invention uses the method combining supercritical fluid chromatographic separation, dual-cation type ionic liquid chromatographic post-column addition reaction and electrospray mass spectrometry detection to establish a novel analysis method for detecting the perfluorinated compounds in a textile, uses a dual-cation type ionic liquid and an electronegative compound to be analyzed to form an adduct having positive charges in the electrospray process, and detects an analyte by means of a positive ion mass spectrometry mode so as to enhance signal sensitivity.
