A method and apparatus for acquiring a high-resolution three-dimensional image of a sample over a range of wavelengths, optionally using polarization sensitive detection is presented. In a preferred embodiment, a spectral domain OCT device is used to sample the complex field of light reflected or scattered from the sample to provide full band imaging. In certain embodiments, structured illumination is utilized to enhance lateral resolution. In certain embodiments, image resolution or depth of field is enhanced by digital refocusing or digital correction of aberrations in the sample. Individual sample volumes are imaged using a single shot technique, and larger volumes can be imaged by stitching images of adjacent volumes together. In a preferred embodiment, a 2D lenslet array is used to sample reflected or scattered light in the Fourier plane or image plane so that the acquired beamlet is distributed over a unique set of pixels in the 2D sensor array.
