Disclosed is a method for quickly measuring the total nitrogen content of urban green land soil by using archaea molecular marker OTU300. The present invention provides a DNA molecule or a probe, as shown in Sequence 1 of a sequence table. Further disclosed is an application of the probe in measurement or auxiliary measurement of the total nitrogen content of soil and an application of the probe in comparison of the total nitrogen content of soil of different plots.
