Methods and compositions associated with using proteins with kynureninase activity are described. For example, in certain embodiments a modified kynureninase capable of degrading kynurenine may be disclosed. Additionally, certain aspects of the invention provide compositions and methods for treating cancer via kynurenine depletion using the disclosed proteins or nucleic acids.
