Provided is a method for preparing a composite microsphere, comprising: preparing a microsphere emulsion, the microspheres in said microsphere emulsion being dispersed in a solvent, the microspheres containing modified or unmodified carrier material and functional particles dispersed in the carrier material; at the same time or after the formation of the microsphere emulsion, applying various physical fields to the microsphere emulsion to control and adjust the distribution of said functional particles; removing the solvent from the microsphere emulsion during or after adjustment to obtain composite microspheres; the multiple physical fields comprise a first physical field used for promoting the functional particles to absorb heat and a second physical field used for causing directional movement of the functional particles. The functional particles in the composite microspheres are distributed in an orderly manner and can be quantitatively adjusted, and the invention can be used in the fields of reversible antibody fixation of DNA fragment separation, DNA recovery, protein purification, in vitro diagnosis, co-immunoprecipitation, cell separation and activation, and single conformation.
