An artificial cultivation method for river-cultivated pearl mussels, comprising the following specific steps: S1, breeding water body selection: selecting a body of flowing water that is distant from industrial areas, receives ample sunlight, and has a surface area of no less than 6000m²; breeding area transformation: S2.1: installing mesh at a drainage location of the water body, and dividing a water source location. By means of transforming a breeding area and diluting the nitrate concentration when supplementing the water of a flowing water source, the present invention makes it possible to prevent adequate sun exposure from being affected by high nitrate content, to prevent nitrate content accumulation from poisoning and killing freshwater mussels, and to prevent the water flow rate from being too high and causing feed or nutritive substances in the water to be washed away too quickly, thus ensuring the growth rate of the freshwater mussels, lowering the mussel death rate and loss rate, providing the freshwater mussels with a better growing environment, ensuring the quantity of pearls produced, and avoiding cost-related waste for breeders.
