The present invention relates to an operation management system and method for optimizing the power generation efficiency of a DMFC applied to an electric car. The operation management system for optimizing the power generation efficiency of a DMFC applied to an electric car may comprise: an electric cart comprising a DMFC, which supplies power by using methanol as a fuel, and for measuring the remaining methanol, power generation and internal temperature of the DMFC in real time; an external server for providing the external temperature of an environment in which the electric car operates; and a data analysis server which updates and stores a learning model for determining whether or not an air filter comprised in the DMFC is contaminated by analyzing remaining methanol, measured power generation, internal temperature and external temperature, calculates power generation by inputting the internal temperature, external temperature and remaining methanol to the learning model, and determines whether or not the air filter is contaminated by comparing the power generation calculated by means of the learning model and the measured power generation in real time.
