The present invention relates to a system for the assembly and modification of nonribosomal peptide synthase (NRPS). The system uses new, well-defined components (units) that include condensed subdomains. This strategy allows for an efficient combination of assembly units called exchange units (XU2.0), independent of their naturally occurring specificity for the subsequent NRPS adenylation domain. The system of the present invention allows for easy assembly of NRPS with any amino acid sequence of choice, without any restrictions due to naturally occurring NRPS units. The system also allows the exchange of natural NRPS components for the invented XU2.0, leading to the production of modified peptides. The present invention provides systems, their individual exchange units, their methods and uses, alongside the nucleic acids encoding these units. [Selection diagram] None
