The invention provides a chemiluminescence substrate having high luminescence intensity, a long wavelength, and good stability, and a preparation method therefor and an application thereof. The chemiluminescence substrate has a structure represented by formula I. In addition, the present invention provides an autofluorescence wavelength and intensity of a chemiluminescence probe in a physiological environment and an application of the chemiluminescence probe in living imaging. Testing results show that the chemiluminescence probe provided by the present invention has autofluorescence of a long wavelength (600 nm), is capable of effectively penetrating a skin tissue, and has high autofluorescence intensity (greater than 10⁷ p/s/cm²/sr), excellent thermal stability (without 1,2-dioxetane structure), and diverse detection groups (applicable to different detection models).
