Live attenuated rabies vaccine is produced by culturing the virus in chick embryo tissue contng. 0.5-5% of a hydrolysed collagen deriv., such as gelatin, which is hydrolysed sufficiently to be dispersible in cold water, but not to the peptone stage. The chick embryo cells are suitably trypsinised before use; are cultured for 1 day; medium is removed; and gelatin hydrolysate -contng. medium is added and cultured till the desired virus titre is reached. The vaccine obtained forms a clear solution in water, is practically free from animal tissue and gives good protection when given to guinea pigs intramuscularly.
