Histidine-rich 3.8 S-alpha-glucoprotein (I) contg. 84 - 4% peptide, 16 plus-or-minus 4% carbohydrate, 9.9 plus-or-minus 0.5% histide; in which electrophoretic behaviour of alpha2-glucoproteins of plasma exhibits an isoelectric point in weakly acid range, esp. pH 5.6-6.2; and having a sedimentation const. of 3.8 plus-or-minus 0.4 S in ultra-centrifuge; mol. wt. 50000-62500; power (a) of binding heparing from which it can be displaced by protamin and (b) to ppte. with a specific antiserum, is prepd. by treating human serum at weakly acid pH with an adsorbent cellulose of heparin covalently bonded to a solid carrier; extracting washed adsorbent with a salt gradient, esp. NH4HCO3; separating first extract peak, esp. by chromatography; concentrating to =2% protein-content; subjecting to preparative zone electrophoresis in a weakly alkaline medium pref. at pH 8.6; separating the alpha2-globulin zone; re-concentrating and opt. purifying, pref. by gel filtration with a molecular sieve having defined exclusion limits.
