1494853 Antilymphocyte Serum INSTITUT FRANCAIS D'IMMUNOLOGIE 18 July 1975 [19 July 1974] 30320/75 Heading A5B Antilymphocyte serum active against human lymphocytes is produced by administering human lymphocytic antigenic material to an animal, collecting the resulting antilymphocytic serum, and purifying it by contacting it with human placental tissue, after which the purified serum is collected. The antigenic material which may be lymphocytes from the thoracic duct or thymus of human beings, is administered to a horse over a period of weeks or months, and then plasma is obtained from the animal. The plasma is preferably treated with 2 - ethoxy - 6, 9 - diamino - acridine lactate at pH 7 to 8 to remove albumens and # - globulins, and then filtered. The placental tissue is prepared by grinding and fixing e.g with glutaraldehyde or formaldehyde, followed by heating at up to 60‹C for 10 hours to inactivate any viruses or bacteria present. The serum is then mixed with the fixed placental tissue, brought to pH 7Œ0.2 and left for about 1 hour with aeitation, and then separated from the tissue. This procedure may be repeated several times until the serum is free from haemaglutinin and haemolysin activity. The serum may then be concentrated by ethyl alcohol fractionation to precipitate an active antilymphocyte fraction, the precipitate then being taken up in buffer at pH 5-6, and dialysed against water. The preparation may be finally purified by chromatography on alumina gel or an ion exchanging cellulose derivative.
