Low concns. of antigens are measured by marking the antibodies with a stable enzyme of high specific activity which retains the major part of its activity after coupling, such as glucose oxidase or alkaline phosphatase, the enzyme being coupled to the antibody with a bridging agents such as glutaraldehyde, and the coupled enzyme is coloured on a plate, the plates then being used to measure antigen concn. corresponding to the marked antibody, by direct or indirect gel immunodiffusion techniques. Method is used in the testing of biological fluids such as cerebrospinal fluid, saliva, urine, intestinal liq., serum etc. The procedure is more sensitive that known immunodiffusion methods.
