Prepn. of N6 -2'-O-dibutyl 8-substd. derivs. of cylic adenosine-3', 5'-monophosphoric acid (cyclic AMP) comprises reacting an 8-substd. cyclic AMP with a butyrylating agent and isolating the product by chromatography on infusorial earth. The products are useful in therapeutic and biochemical research for stimulating the synthesis and/or release of certain hormones by the endocrine glands. The process gives yields 80% and is suitable for large-scale use. The 8-substituent is pref. SH, Br or MeS. The butyrylating agent pref. comprises butyric anhydride and excess anhydrous pyridine. The reaction is pref. effected at room temp. for 5-8 days, opt. after refluxing for 30 mins. The infusorial earth is pref chromatography-grade kieselguhr.
