Prodn. of gamma globulin solns. from intravenous injection - by treating a normal globulin soln. with e.g. ethyl acetimidate hydrochloride

Prepn. comprises treating a normal soln. of gamma-globulin with (per g of protein) (A) 0.055g ethyl acetimidate hydrochloride; (B) 0.022g diketene; (C) 0.05g ethyl formimidate hydrochloride or (D) 0.077g propane sultone. Treatment is at 5 degrees C for (A), (B) and (C) and at 37 degrees C for (D), and at pH 9, and after allowing to react for 2 hr. the pH is adjusted to 7-7.5 then the mixt. dialysed and sterile-filtered. The parent patent described the modification of gamma-globulin with other alkylating or acylating agents such as beta-propiolactone. The compsns. have specific complement-fixing activity so cause bacteriolysis in cases of bacterial infections. In cases (A) and (B) reaction is more rapid than with propiolactone so can be carried out under milder conditions; (D) introduces sulphonamide gps. into the protein which themselves protect against infection and in (C) the mol. wt. of the treating agent is so low that only a minimal alteration in the immunological properties of the globulin occurs so tolerance is improved on longer-term administration.
